i have not done this on vista, but try this.
right click on the directory -> properties -> security -> edit -> add -> Advanced -> Find Now
Add your user, and make sure IIS_IUSERS, IIS_WPG, and whoever else needs access to that directory are added.
Just a guess based on how I would troubleshoot IIS on previous versions of Windows.
